Halorarius litoreus gen. nov., sp. nov., Halorarius halobius sp. nov., Haloglomus halophilum sp. nov., Haloglomus salinum sp. nov., and Natronomonas marina sp. nov., extremely halophilic archaea isolated from tidal flat and marine solar salt

Five novel halophilic archaeal strains, named BND22 T , ZY10 ZY41 ZY58 and ZY43 were isolated from the coastal saline sediment of intertidal zone located in Qingdao natural sea salt produced Huanghua marine solar saltern, PRChina. These five strains demonstrated typical morphology growth characteristics haloarchaea. The comparison 16S rRNA gene revealed that strain BDN22 was associated with Salinirubellus salinus ZS-35-S2 (95.2% similarity), related to Halosegnis rubeus F17-44 (95.1% both closely Haloglomus irregulare F16-60 (98.1% 98.2% similarities, respectively), close Natronomonas salina YPL13 (98.0% similarity). In addition gene, rpoB′ is undoubtedly another important molecular marker for identification archaea, concatenated-conserved-protein phylogeny widely used classification recent years. Phylogenetic phylogenomic analyses based on genes 122 concatenated protein showed these haloarchaea construct three different clades gathered together current members Haloarculaceae . Strains formed two distinct separated members, clustered Natronomonas. ANI, DDH, AAI values against phylogenetic neighbours no more than 91%, 45%, 92%, respectively, far below cut-off species delineation, supporting their placements new taxa.
